One-Pot Consecutive Reductive Amination Synthesis of Pharmaceuticals: From Biobased Glycolaldehyde to Hydroxychloroquine

Reductive amination plays a paramount role in the synthesis of amines. It is often proposed as more ecofriendly process than traditional SN2-type reactions amines it avoids toxic alkylation reagents such alkyl halides. This work demonstrates versatility reductive reaction via hydroxychloroquine (HCQ), one most renowned pharmaceuticals during this coronavirus pandemic. The novel green strategy based on three consecutive conducted one-pot system, avoiding intermediary purification steps. Furthermore, biobased C2 platform molecule, glycolaldehyde, was selected starting reagent. newly developed pathway appraised using CHEM21 Green Metric toolkit and compared with commercially operating method.

A one-pot, reductive amination/6-endo-trig cyclisation for the stereoselective synthesis of 6-substituted-4-oxopipecolic acids.

The first stereoselective synthesis of 2,6-trans-6-substituted-4-oxo-L-pipecolic acids using a tandem reductive amination/6-endo-trig cyclisation process is described. The sequential reduction and cyclisation mediated by sodium cyanoborohydride allowed the preparation of a series of highly functionalised 6-alkyl and 6-aryl analogues.
